Once you get to the top of the fortress, you have to deal with wave upon wave of zombies as they swarm from the Occult General. Don't kill the general yet.
Before the two sets of stairs that lead up to the general is a mounted machine gun, just man that and mow down all the zombies. They spawn endlessly but in distinct waves.
At the end of each wave, dismount from the gun, and double check there are no injured, crawling zombies. I died because I did not look out for them.

This will come naturally as you progress through the campaign and Horde mode. It doesn't matter what method you kill zombies with and all types of enemies count towards the trophy. You will receive periodic progress updates towards this trophy as you play the game. You can also track your progress by navigating to Extras => Campaign Stats from the main menu. If you are adamant on grinding this trophy, then you can play any Horde mode in solo mode and set the enemy spawn to 'For 4 players' and the difficulty to Cadet. This will drastically increase the number of enemies and as you are playing on your own you will obtain all the kills. All kills count towards the trophy regardless of game mode or if you die at any point.

Just kill 5,000 enemies of any kind over multiple playthroughs; skeletons, zombies, suicide zombies, elites, demons, summoners and summoned zombies all count toward this. I'm not sure if fire zombies that demons create count though.
