You need to max out fame in challenge mode to unlock all the animals, and possibly complete some of the campaign scenarios. I think for this achievement you have to research 256 different items. As I have been playing through the campaigns, I have noticed that my progress bar doesn't seem to be progressing unless I am researching items that I haven't researched before. I feel like I have definitely researched more than 256 total items so far since I have done all the campaigns and a few challenges, but my completion percentage is only at about 70% .

I'm max level 40 and I have not unlocked half of the animals. I'm not sure fame matters much. I think it's based on finishing up other animals to get more animals to unlock, hence more to research.
