The Vampire character is unlocked by getting 40% life steal in a run. Then play a run with Vampire and win it. You can use the difficulty sliders to make it easier.
To get the 40% life steal, use the Sick character that starts with 25% then focus on getting items like Teeth that increase it. Reroll as needed when it’s cheap to do so to increase your chances to get these life steal items. You get the Sick character by getting -5% health regeneration in a round - focus on grabbing upgrades that decrease it to get that.

I can't remember, but once you hit 40% and unlock the character you can just end run?
Edit: Yes, once you unlock the achievement and new character you can return to menu and select them.

The vampire character is an odd one to play with. Requires constantly taking damage - but not too much! - and then backing off to kill things. I found that in the rounds with tougher enemies, it was best to kite everything until there were a decent number of big guys, then take about half damage and unload on them. Fun stuff.

This character wants you to lose health constantly to make it have more damage. However, you can just get stat upgrades to offset the starting damage deficit and then eventually just play like normal, which is what I ended uo doing.
