How to unlock the Others and Wights achievement in State of Decay 2: Juggernaut Edition - Definitive Guide
The fastest ways I've found to kill zombies are fairly obvious - run them over with a sturdy car and antagonize plague hearts (although this is very dangerous). Right at the top, know that kills your followers get do not count, nor do friends kills count in co-op. (EDIT: Since this isn't clear, this is cumulative over all playthroughs you have in State of Decay 2.)
Running them over
Running them over is the safest bet, but it takes more work. The best way I found would be to have the auto lot in a large slot on your base, upgrade a medium size car and then mowing them down as you have tons of repair kits and fuel canisters. As ProfessorX 86 said, make sure to keep slamming on the horn or have the sirens rolling (on an ambulance or police car) to get zombies around you to come in. If you upgrade vehicles with sirens, however, the siren is removed (Thanks Luna go sky and AES0P). It has also been mentioned in the comments (thanks again AES0P) that the Auto Shop has an ability to reduce car damage for an hour.
EDIT: In addition to this, Pulzy 21 brought up some good points I forgot to mention about the car idea. You want to make sure that you have a steady stream of parts and materials for this. Parts help you make repair kits with a fully upgraded workshop. These are the key to the car idea (gas cans are easier to get than repair kits in the wild). Ideally, a base would have a workshop or a forge, and an auto shop as a base to make sure you get the most out of the car idea. Breaking down materials rucksacks also net you 30+ parts a shot and I've noticed that seems to be the better bet when your base is already at 30+ materials and you just need parts fast. There is also a slight chance that breaking apart the rucksack will net you a repair kit. Good luck!
EDIT: Zarda1 had a good idea for those without sirens - remap the horn to a button that is easier for you to hold down. The accessories app on Xbox should allow you to remap. Also, for those with an Elite controller, I'd advise putting it to a paddle.
DLC CONTENT EDIT: With the introduction of the Independence Day Pack, I thought I'd mention that the three new vehicles - Pyrohawk (a hatchback that shoots off fireworks), the Burninator (a truck with side-mounted flamethrowers), and the Meatwagon (a cargo van that drops delicious red meat) will all greatly help kill and attract zombies. The pack is free for this with the Ultimate Edition, but $4.99 USD otherwise.
SECOND CONTENT EDIT: Dr Scruffleton is reporting that the Church on The Hill base available on the hills map (Cascade Hills) has a bell tower that constantly attracts zombies to your base. Driving around blaring the horn is apparently a nice way to attract zombies to your location at the base. Whitney field Base in Meagher Valley has the same effect (thanks AES0P for the hyper passive-aggressive comment).
DLC Update - Daybreak: User Skeletaidonut had this to say about Daybreak: "For anyone reading this the Daybreak DLC counts towards total killed and if you complete all 7 waves you can get upwards of 700 kills a match."
PERSONAL EDIT: I was able to kill 3.9k zombies in 4 hours and 25 minutes. So, it is possible to kill that many zombies in an hour, but I also have a follower with the "Driver" trait that gives 100% fuel efficiency and 30% impact reduction. Highly recommend finding that trait if possible and having an auto shop roll the 30% impact reduction as well.

FINAL EDIT: As more DLC's come out, be sure to check the comments as the most up to date information could be in there. In general, this achievement is best won via running over zombies in any of the modes. Horde mode gives you more chances, but you can naturally get this in your playthrough. @Dr Scruffleton I've finished this achievement and 1000 in an hour driving around seems pretty optimistic from my experience. Also as you've noticed the plague hearts don't constantly spawn zombies, they spawn them based on the damage done to them. I went in with the 50 cal Timberwolf and shot it twice to antagonize it hoping zombies would keep spawning, it would only spawn 20-30 zombies and then it stopped for me. The only benefit I can see to the plague hearts would be leaving them and all infections untouched, then circle the map in your vehicle to try and let the hordes pile up around each while your making laps. Another thing I noticed but haven't tested (and not going to bother testing now) is that there are at least 2 bases that have a unique facility that can call in zombies. The Church on the Hill in Cascade Hills has the church bell and Whitney Field in Meagher Valley has the announcer box which can both be used to call in hordes of zombies, in case anyone wants to test those and report back for others.
Also Dislodged Puma, you may want to add to the solution that the auto shop also has an option to decrease damage to vehicles for an hour, which can be useful if your using the driving method.

Ultimate edition DLC has an amazing car for this! I recommend starting a new community, with the builder and trader boons, which you should have already, because there's no reason going for this achievement if you don't have all others.
so, to clarify:
Make a new community
Select Trader and Builder boon
Fill 2 characters from previous communities if you want.
Make sure you reroll a random survivor until you get the Driver skill. This will greatly reduce the amount of fuel and repair kits that you will use. (autoshop adds another 30% that stacks)
Call in the ultimate edition vehicle if you have it, costs 400, but you start the new community with 4000 because of the selected boons.
Go ham and run over everything.
I also personally recommend your base as the football court in Meagher Valley, as you have good base defenses here and can attract a lot of zombies. And what fun is a zombies game if you keep them all away from your base right?

For this achievement I made sure
-Choose trader as leader/builder boon from previous playthrough
- choose Meagher valley
- Get Squelones brewery asap (1500 influence, 6 survivors)
- Use 1 large slot for trading post
- 3 small slots for hydrophonics, 1 workshop,1 infirmary, and the other large slot auto shop
- Make influence from brewing packs of crafted beer (cost 3 food but with 3 hydrophonics and using seeds on them you should be pulling 16-20 food a DAY in)
- call traders for ANYTHING (this is were the trader leadership comes in handy- the ability to call traders to your base assuming you’ve made the TRADING OUTPOST) your missing, supplies!!, get a CNC saw installed (again bought from traders/found in the world) and convert supplies to parts for your TOOLBOXES -you buy all your FUELBAGS from TRADERS and use the storage icon to convert into actual fuel for your vehicle. use the 3-4 outposts for bed slots, or whatever you want.
It’s upto you what vehicle you want to use but I’d HIGLY recommend the Maximilion, get a medium vehicle upgrade kit on it to make it the MEGA MAX (which can be bought from calling a parts trader to your base) I say this because I tried using the road racer etc and I found the handling very stiff, with the maximilion you can pull on the stick and whip the car round better than any other vehicle I’ve come across - also I’d put the horn in the settings to an easier default button, I chose ‘B’ which was the handbrake (won’t need it in maximilion)
Anyway use your self efficient running base to craft all the supplies you need whilst your out on road runs smashing zeds - fill the trunk up with fuel and your invent with tool boxes and 1 fuel can i was using 2-3 tool boxes per fuel canister - I feel like I’ve rambled on a lot, one last thing if anyone reads this message my GT I’ve lots and lots of spare tool boxes, gas cans and influence (10k+) so ill stock you up as much as I can 🤗
