Did this last night and it worked like a charm!! A friend of mine told me about this and thought I would post it for all.
Set up a player match with you and a friend.
The person going for the kills becomes an engineer.
Build a dispenser first then use the money out of it to build a sentry right beside it. Make sure you upgrade the sentry by hitting it with your wrench to its max.
The other player becomes a spy and needs to disguise himself so the sentry does not kill him.
Go up to the sentry and place a zapper on it - The Engineer hits it with his wrench and it will count as a kill. Rinse and Repeat. If you are close enough to the dispenser you will regenerate money also so you will never lose health on your gun or Engineer.
The kills will not count in your stats until the Engineer is killed by the spy. Me and a buddy of mine got about 1800 kills in about 1 hour and 45 mins and we had some "goof off" time in there.

Now that the game is backwards compatible you can get this achievement by yourself if you are fortunate to have a 360 and an XBOX One.
Once set up, rubber band RT on the engineer and use a sapper every few seconds on the other account.
Use your live account on the 360 and another one on the one as you can share gold on that !!!

There is an easier way than these other solutions, and will stop you accidentally being killed or messing around with disguising yourself due to the sentry turret. This was my idea and worked a treat in a gaming session a couple of days ago. Start up a game as normal, and then one person selects a spy and one person selects an engineer. The engineer sets up a dispenser unit. ONCE THE UNIT IS BUILT the spy can put up to 4 sappers on the dispenser in a row while the engineer whacks them off with his wrench. Make sure you do not do this early as otherwise the dispensers health will still be low and you run the risk of breaking it and then having to kill the engineer so he will have enough materials again. After this, the health gets too low and you have to wait one second (literally). After this, the health will be full and free for another four sappers. For each sapper smashed, the engineer will get one kill. These kills are then counted ONLY when the engineer is killed. This method literally gets you just shy of 30 kills a minute if you do it right. (I timed it exactly). If the dispenser is accidentally broken, set another up and try again. The engineer can also hold or rubber band the right trigger to hit the dispenser with his wrench. The spy takes a bit more careful timing. For each hit made to the dispenser itself without a sapper, the engineer will increase materials in case of mistakes.
EDIT 05/02/2012: To secure the shortest time taken to get this achievement, (particularly if you are worried about making the mistake of destroying the dispenser early or the engineer running out of materials - this happened to me about 3 times total whilst aiming for this achievement, so it does happen!) instead of killing the engineer to get him more materials, you can use this method near the engineers spawn point so he can run in there and replenish his materials, therefore avoiding long respawn times (for mistakes) or just cutting down on steps taken to return to the boosting destination. (credit to Quarantane for this helpful tip/warning/alternative).
HOWEVER - you must remember to kill the engineer every so often because (as a reminder) THE DISPENSER KILLS WILL NOT COUNT UNTIL THE ENGINEER IS KILLED. This meant that any mistakes that I made were almost welcome, as it was another chance to secure my kills. If the game time runs out and the engineer has not been killed at the end, he will lose all kills since he last was.

Just to point out, this is essentially the same idea as the sentry solution, you're just using a dispenser instead to avoid "messing around with disguising yourself" which takes very little time, and can be finished by the time you walk to the dispenser you're going to sap.
And instead of killing the engineer to get him more materials, you can just use this method near the engineers spawn point so he can run in there, to avoid waiting for a long spawn time.

Nice point about the engineer's spawn point. Will add that to the solution with credit to you. Also I had problems with being killed by the sentry turret and it got very annoying very quickly, but the dispenser is no risk, and therefore much easier in my opinion. It was very well appreciated by everyone in the boosting session I attended, so it is a worthwhile solution.

All this requires is time commitment. There are multiple ways to do this but first are the no-no's. Do NOT use the medic, they are awful offensively. Also, unless you are good at backstabs, the spy is a bad choice as well. I used the pyro and soldier the most. If you use the pyro, flame all of your teammates when you see them, they may be a spy and you can get a free kill. An engineer may also work well if you put your turret in the right spot. On 2fort, I find the best places are at the top of the outdoor room with the 2 staircases and in the intel room in the corner. If you are patient, you will get this achievement eventually.

Simply get 1000 kills on Team Fortress 2. You can play player or ranked matches for these kills. Depending on what character you are good with, you will get this much faster than others
I prefer the Engineer on the 2fort level, just set up a sentry, upgrade it, and scout out surrounding areas for enemies and lead them into your sentry. I usually pulled out about 30 kills per match that way or I used the scout and just ran circles around people, lots of fun :)
Other classes for high kills are demoman, soldier, and heavys, all of which when paired with a medic are killing machines (especially when they get ubercharged). Also if you are a good sniper you can pull off several kills very quickly.

FacelessEnd and I were attempting WolfVelocity's method, which I know works very well, but in setting up a private and a ranked match we were unable to spawn as spies. We tried this two different days and no luck. After that, we tried sageon's method, and although it does work, it is a bit on the slow side. While we were plugging away with sageon's method for our second boosting session, I developed a method that is faster than sageon's method and nearly as fast as WolfVelocity's (a good substitute if you can't spawn as spies).
1. Play on 2Fort
2. Both play as Engineers
3. Each players build Teleporter Entrances across the room from their base's garage door spawn room (NOT near the doorway, as explained later) and then run across to the other side and build a Teleporter Exit beside the other players Teleporter Entrance.
4. One player (Player Y) will stand near the garage door of the other team. The player (Player X) who is at their home base will build a sentry gun (which is then immediately destroyed by Player Y) and then Player X will run into their spawn room to the lockers, refill money (automatic) and run out the door to quickly build another sentry (to be destroyed by Player Y).
5. Once both players get in the rhythm of things, Player X will be able to run backwards to the lockers, saving time for refills, and Player Y will be able to use 1 pistol shot on each sentry gun as it is being created. I recommend building sentry turrets, because when you bring up the building plans it is the first on the list, and easiest to select with minimal button presses.
Note: *This is why I say build Teleporters!*
The achievement won't pop (i.e kills don't register) until the player who is doing the destruction gets killed. Every 10 minutes or so Player X should kill Player Y so that he can check on his achievement progress. The Teleporters quickly get you back into the action.
FacelessEnd and I were both able to get 180 to 200 "kills" every interval of approximately 10 minutes once we got into the rhythm (which doesn't take very long despite how complicated this sounds). We destroyed turrets until we ran out of pistol ammo and then died as our method to keep the best tally possible.

The way I did it was with a friend of mine in a private match. both of us were engineers, we both built a dispenser close to where we were and filled up our medal then went about building teleport entrance/exits (both) and as the building process went by we would destroy them and we would just keep doing that. once we got into a groove the kills just kept growing we got about 400 in about 40 minutes. its quicker than the spy and engineer and plus both would be getting it. once you think you reached 1000 then just kill each other and check your progress. (That was just with two people i'd imagine it be even quicker with more people) Good Luck!
I can do it better than the rest of these solutions. This will allow any number of people to get 450 kills every 15 minutes, EACH.
The number off players does not matter. You could have 9 people and this will work. Really, the more the merrier.
You do a similar strategy to the other solutions, with a spy placing sappers on a turret. Here's how you set it up:
1. Spy stands in middle of the room. Engineers build turrets around him a circle. They then build a dispenser behind them so that it looks like this -

3. The engineers face their turrets with their wrenches and hold R. Then press the guide button. This will make your action continue forever. You can now get up and leave the room.
4. The spy needs to make sure his look sensitivity is set to default. If it is too fast this will not work. Default works perfectly. Set it to default.
Hold right on the right joystick and hold R at the same time. Then press the guide button.
The spy should now be placing a sapper on each turret very quickly, and the engineers should be automatically taking them off.
Conclusion:
It took us an hour to get 3 people from zero kills to the achievement. We didn't have to put any effort into it and quite a bit of time was spent figuring out how to get this to work.

Ok.. going to add another slice of good cake to WolfVelocitys notes (we did test I8ITackyticsI8I method but it didn't seem as fast as the following)

Host a player match, Set the game mode for 2fort, unlimited time, no autobalance and 8 private slots. party up on xbl with either 1 or 2 friends and invite them to the game
1. Do exactly as WolfVelocitys says, however as soon as you start building the turret, hold down the right trigger and tap the xbox guide button, then let go of the right trigger. For some reason the engineer will keep repairing the turret and you don't even have to touch the controller.. as long as the spy doesn't load up the turret too quickly you're golden (don't forget to nugde the stick everyonce in a while if you're on battery).
2. In a 3 player game you can do the same again, 2 engineers vs one spy. both engineers set up a little distance from each other, within reach of the Spy. Both engineers set their controllers to 'auto fix' (as above). The spy stands in the middle holds left or right on the right stick to rotate (not full speed though) whilst holding the right trigger, which will automatically set the zapper and allow enough time for the engineer to replace. Theoretically with a bit of testing you could rubber band this method. The spy should also hit Y to keep a tally of the kills.
Last method for 1 vs 2 boosting:
same as above, both engineers set up dispensers and turrets, but place the turrets close together. One engineer fixes both turrets whilst the spy places zappers. The other engineer is on standby in case the one of the turrets looks like it might blow.
All of the above are fast ways to boost. We obtained around 1600 kills between 3 of us in about 1.5 hours last night.

This is an addendum to WolfVelocity's solution for those wondering why they can't pick a Spy to do this with.
This has definitely been halfway patched. I tried a dozen different ways of doing system link, player match, etc using my two consoles and it would NOT let me pick a Spy with only two players (myself and my dummy) in the match. As soon as I deleted the title update by clearing my system cache, I was able to pick a Spy with my dummy account.
My guess is you need at least 3 or 4 players in the match before it will let you take the Spy class if you have the title update installed, so just do this offline until you get close to 1000 then install the title update before you finish it off.
The only pain in the ass part about this is that if you're using the games-on-demand version to be able to run two xboxes with only one purchased copy of the game, you'll only be able to play about 9 minutes at a time. This is because you have to download the title to your main console, which will then have the 'digital rights' to the game, then move your main gamertag to the secondary console and download it again.
In order to be able to play the game AT ALL without being connected to xbox live, the console in question needs to have the 'digital rights' to the title.
So the original xbox you download it on will be your dummy account and will be able to play it without your main gamertag signed in. The secondary console you will be using with your main gamertag, but since you have to delete the title update you won't be able to stay connected to xbox live.
You launch the game while connected to live and it disconnects you for the title update and starts a 10-minute countdown timer. At the end of this timer, if you haven't downloaded the title update and connected to live to verify ownership of the game, it kicks you out to the dashboard.
By the time you start a system link game, get your engineer and Spy set up, and get going, you're left with maybe 8 minutes left give or take.
At 30-ish "kills" a minute by destroying the sapper, you're looking at 240 kills give or take before you have to kill yourself before the timer expires (you do get a warning every minute of the countdown) to get the kills to register. Just after the one-minute-to-go warning, kill yourself, making sure your kills registered in the achievement progress, then dashboard out of the game and restart the process.
I did this 4 times total, making sure to stop when I was within 10 kills of the 1000-kill mark. I then downloaded the title update so I could stay connected to live for the last few kills to datestamp the achievement.
I hope this helps people wondering why it won't let them take a Spy.

I would like to give credit to my friend zzDazmanzz for discovering this.
There is a glitch you can do to get this achievement, and it's a good way for the Engineer, but not for the Spy.
1. Create a Private Match and invite your friend.
2. The player who is going for the kills chooses the Engineer, the player who isn't is the Spy.
3. Meet up on the map and get the Engineer to build a Dispenser.
4. Ensure the Dispenser is fully upgraded. You may need to kill the Spy once or twice to get it to work.
5. The spy may need to change it's outfit to the opponents coloured Spy.
6. The Spy needs to constantly place a Zapper on the Dispenser, and the Engineer needs to press and hold the Button to whack the Dispenser. This will keep it upgraded.
7. The Glitch: Prepare to see a Hilarious outcome for the player who is the Engineer. Whilst you are holding down the Button, quickly press your Button. If done right, the guide will open but the Engineer will still be whacking the Dispenser. Keep your XBox Guide open and the Engineer will keep on whacking the Dispenser so you don't have to hold down the Button anymore. You can look at your achievements friends list or other interests.
Note: Don't press the Button a second time, or the Button on the Main Guide Page, as that will stop the Engineer from whacking the Dispenser, and you'll have to retry the glitch again.
Sadly for the Spy the glitch didn't work, so they will have the distressing part of pressing on a regular basis to place a Zapper.
8. When you want to find out how many kills you have done, get the Engineer killed by the Spy and it will update your Stats, when you press the Button. If you have met the requirement of 1,000 Kills, but you have not got the achievement, simply get yourself killed by the Spy and the achievement will unlock shortly after your dead.
This method should give the player who is the Engineer, the achievement in around 1hr 30m.

8. When you want to find out how many kills you have done, get the Engineer killed by the Spy and it will update your Stats, when you press the Button. If you have met the requirement of 1,000 Kills, but you have not got the achievement, simply get yourself killed by the Spy and the achievement will unlock shortly after your dead.
This method should give the player who is the Engineer, the achievement in around 1hr 30m.

How to get this achievement in Solo play WITHOUT Xbox Linking or Cross-Console play
This method takes some time.
Host a Game in System Link Match on the map Dustbowl.
Some barrels in Dustbowl count as kills, there are 10 barrels in the map, 7 are destructible and count as kills. There are 3 destructible barrels in Stage One, 4 barrels in Stage Two and none in Stage Three. Kills are added to the achievement at the end of a Stage, so complete Stage Two, when Stage Three starts, pause and hit disconnect and try again!
This method takes some time.

I would like to share a few tips that helped me.
1) You can set up private slots so that random players don’t annoy you.
2) While playing as an engineer, you can hold down and press while doing so. Your character will stand afk and you will not need to touch your controller.
3) When playing as a spy, install Sapper a little earlier when the rocket silos on the engineer’s Sentry Gun is equal to the horizontal position - 180°.

One easy way of achieving this is to set up alot of turrets in every game. You should get it pretty soon if you have a fully upgraded turret on a map picking off enemies. This can be achieved in Player Matches with friends.
