Uncle Fester's guide really covers just about everything you need to know. There are many different group and trinket compositions that will get you the achievement (see the comment thread below Uncle Fester's Guide). Instead of repeating what was put there, I'll simply relay my personal experiences that (finally!) worked in trying to get this achievement.
My team:

My trinkets:

Vestal - Sacred Scroll (better healing, lower stress), Heretical Passage (better healing)
Jester - Bright Tambourine (lower stress taken, better stress removal skills), [anything else -- I didn't have much]
Shieldbreaker - Heavy Boots (protection, move resist, lower speed), Spectral Speartip (increased damage and HP)
Sheldbreaker - Fuseman's Matchstick (increased DMG and critical chance on ranged attacks), Hag's Ladle (increased blight chance)
NOTE: the Heavy Boots on one Sheildbreaker was to try and get that Shieldbreaker to go second in any given turn, thus enabling me to use "Impale" or "Adder's Kiss" with each Shieldbreaker in succession (see below).

Camping Skills my characters used:

Vestal - Pray, Santuary (both stress relief)
Jester - Every rose has its thorn (stress relief), Turn back time (stress relief), Tiger's Eye (accuracy and critical chance boost for a sheildbreaker)
Shieldbreaker - Snakeskin (protection and HP boost)
Provisions:
------------
Food, antivenom, bandages, medicinal herbs, keys, holy water, laudanum -- as much of these as possible. Holy water is optional, but I needed it for the added trinket boost to healing for the Vestal
Do not bring: torches (there's no need), shovels, and other DLC provisions, including shard dust.
General Character Strategy:
------------------------------
Jester -
1. Battle Ballad most of the time - increase party speed, accuracy, and critical chance; plus buff Finale's damage and critical chance, should it be needed
2. Inspiring Tune - decrease one party member's stress, plus buff Finale just like above
3. Finale - on bosses, or strong opponents, but only when the opponents had re-arranged my squad resulting in the Jester being in the front (the only place from which the Jester can use Finale)
4. Dirk Stab - used very rarely, but good to finish off baddies or move the Jester forward
Vestal -
1. Switch between Divine Grace to heal one person, or Divine Comfort to heal the entire party. NOTE: a critical heal on a character will often (always?) reduce their stress!
2. Judgement - used when I want to do a bit of damage, and also heal the Vestal
3. Dazzling light - used to stun when I thought I could get away with it
Shieldbreaker-
1. Impale - used when there are multiple enemies and the Sheildbreaker is in the first position. Spreading blight among the baddies is great. Also keep in mind that this hits all the baddies - meaning you have four chances for a critical, and remember that the critical hit has a chance to do some stress heal to you and possibly a party member.
2. Adder's Kiss - used when in the front spot, and when a larger foe is in range. 5 points of damage over time is pretty nice
3. Pierce - used when the Shieldbreaker gets a turn and isn't in the front spot. This attack is armor piercing.
My typical turn would be: Jester (Battle Ballad or Inspiring Tune, as needed), Shieldbreakers (Impale or Adder's Kiss, as dictated by the situation), Vestal (heal). And that's really it. Rinse. Repeat.
Tips:

I've put some in the text above. However, some advice bears repeating. If possible, use Sheildbreaker's Impale -- this can help with stress healing. Likewise, use Vestal's Divine Comfort for the same reason -- critical heals can relieve stress.
The Vestal is there to physically heal, rarely to do damage or try for a stun.
The Jester is there to speed up the party and stress heal, rarely to do damage.
The Shieldbreakers are there to infect everyone with blight. The armor piercing is nice, too.
Beyond that, the random stuff that can happen to your squad can hopefully be manageable. This isn't different from the rest of the game, but multiple boss fights can really wear down your party and make a run to 300 impossible. I personally think several of my runs were ruined by some bad luck -- just like some runs in normal dungeons. This is Darkest Dungeon, things can turn on you quickly and become exceptionally frustrating. However, with the above crew, trinkets, perseverance, and a bit of luck, I got this achievement.
Please let me know how I can improve the guide if you have a differing opinion. I merely noticed the lack of guide and wanted to at least put something up. I highly recommend giving Uncle Fester's Guide a read -- there is a ton of good material there.
My shield breaker had adders kiss, impale, pierce and the forward one. I started off without adders kiss for a time and had the shield but changed my mind at 200 in or so.
I had a sunken crew around 250 which nearly got me but a shambler at 290ish was just fine. Good luck all
300 kills is the hardest trophy in this set. You will be at approximately 100 after defeating the Sleeper and then you will get a random boss fight at the end of each bar. The main issue with this is stress so a Jester is very important to help with stress mitigation and buffing. Another issue if you are trying to go a long way is diseases so might be beneficial to bring a Plague Doctor or Graverobber to remove diseases. The team I used was Vestal - Jester - Man-at-Arms - Leper and I made it to 300 kills easily by abusing the Splendorous light and virtuous breaks. Vestal and Jester are very important for this, but the other 2 can be swapped for what you prefer. Man-at-Arms worked pretty well for his buffs and being able to hit the 3rd slot enemy and Leper for being able to focus the front 2 pretty hard.

1 and 2 - Shield Breakers
Because The Vestal is the best healer, the Jester is the best at relieving stress and buffing the team, and if necessary, she simultaneously hits the 2nd and 3rd positions of the enemy with bleeding. Well, Shieldbreakers are great at handing out cradles, reaching any position from almost any position, etc.
Also not an unimportant point: before entering, rid the entire squad in the sanatorium of negative character traits and diseases.
Regarding decorations:
Vestal - to increase healing skills
Jester - to increase stress relief
Shieldbreakers - of course for damage.
At the same time, if there are no suitable tricknets, then they will also be useful against poisoning; I almost never encountered bleeding in all my attempts.
I also advise you to try not to waste antidotes and bandages, but to heal the damage with the Vestal, save your supplies for the most extreme case!
I set the following skills, I’ll chew them just in case:
Vestal:
- Condemnation - hits any position, weakly, but hits.
- God's Grace - heals one selected character well
- God's consolation - average healing for the entire squad
- A blow with a mace - it just hits, I hardly used it, incl. You can choose any other skill.
Jester:
- Strike with a dagger - hits from any position + moves one position forward.
- Harvest - hits 2nd and 3rd positions + inflicts bleeding.
- Battle ballad - strengthens the entire squad: +5 accuracy, +2% to critical chance. hits, +3 speed / Hero: final: +20% damage (7 rounds)
- Invigorating melody - relieves stress from one selected character
You can, of course, replace the Dagger Strike with the Finale, but this is not for everyone.
Shieldbreakers:
- Penetration - usually a very strong blow to one enemy + moves forward
- Stun - medium hit on one enemy, plus pull + moves forward
- Pierce - medium attacks all enemies + chance to inflict poison + moves backwards - THE BEST STRIKE!!
- Piercing - hits one player + moves backwards.
As a result, the battle looks like this:
The Vestal Virgin only heals. Attacks only when everyone is completely healthy.
The jester only relieves stress. It buffs the team only if everything is ok with stress. Attacks as a last resort.
Shieldbreakers: - exclusively attack! At the same time, try to hit with the Piercing skill - damage to all enemies + poison. Acts only from the first position, and after being hit with a shield, it jumps back one position.
What’s the fun: Once you understand the trick with moving the Jester (Dagger Strike) and Shieldbreakers after each blow, taking out enemy units in batches will not be so stressful.
IMPORTANT!!! The character has become psychotic?! This does not mean that the campaign is a failure! If you understand that you can reduce his stress from 100 to 0, then the psychosis WILL GO AWAY!! And you can safely continue.
PS: But remember, the terrible god Random is great, treacherous and dangerous!
Played by a vestal jester and 2 shieldbreakers. Plus, everyone was wearing -20% stress relief books, but that didn’t help much. I came across 2 stressful bosses in a row, when the counter was already 250, and the poor heroes were already ready to die from stress.

PS I played it on Darkest difficulty. I don’t know how much easier it is in light mode.
